Photoreceptor Phytochrome Regulates the Cyclic Guanosine 3′,5′-monophosphate Synthesis in Avena Sativa L. Cells

چکیده
The activity of guanylyl cyclase (GC) responsible for cGMP synthesis was found to be predominantly located in a fraction of plasma membranes in cells of Avena sativa seedlings. The GC activity was shown to depend on light conditions of plant cultivation and seedlings age. The reversible regulation of the GC activity in etiolated seedlings by red/far red light was found. This suggests that photoregulation of GC activity is mediated by phytochrome in cells of oat seedlings.

In attempt to elucidate the early molecular events associated with the biological action of cyclic guanosine 3/,5/monophosphate (cGMP) its binding to the structural components of Avena sativa L. plant cell has been studied. cGMP was shown to be bound specifically to proteins located predominantly in the soluble cytosolic fraction. cGMP-agarose affinity purification procedure followed by SDS-PAG...

Phytochrome A (phyA) is a red/far-red (FR) light photoreceptor responsible for initiating numerous light-mediated plant growth and developmental responses, especially in FR light-enriched environments. We previously showed that the first 70 amino acids of the polypeptide contain at least two regions with potentially opposite functions (E.T. Jordan, J.R. Cherry, J.M. Walker, R.D. Vierstra [1996]...
متن کاملInhibition of phytochrome synthesis by the transaminase inhibitor, 4-amino-5-fluoropentanoic Acid.
Gardner and Gorton (1985 Plant Physiol 77: 540-543) demonstrated that the transaminase inhibitor gabaculine (5-amino-1,3-cyclohexadienyl-carboxylic acid) inhibits the initial synthesis and resynthesis of spectrophotometrically detectable phytochrome in vivo. Another mechanism-based transaminase inhibitor, 4-amino-5-fluoropentanoic acid (AFPA), is examined in this report for its effects on phyto...
متن کاملInhibition of phytochrome synthesis by gabaculine.
Gabaculine (5-amino-1,3-cyclohexadienylcarboxylic acid), a transaminase inhibitor, also inhibits chlorophyll formation in plants, and the effect of this compound can be counteracted by 5-aminolevulinic acid (ALA) (Flint, personal communication, 1984). Since it is probable that ALA also serves as a precursor to phytochrome, the effects of gabaculine on phytochrome synthesis in developing etiolat...

The amino acid sequences of phytochrome from Avena sativa, Oryza sativa, Curcurbita pepo, Pisum sativum and Arabidopsis thaliana have been analyzed with a variety of computer programs, with a view to identifying areas of the protein which contribute to the properties of this photoreceptor. A region at the C-terminus has been shown to be amphiphilic, and by analogy with surface-seeking peptides,...
